Decreased inflammatory response in rat bladder after intravesical administration of capsaicin-loaded liposomes Anais da ABC (AABC)
CIRINO,LORENA M.D.; VERGNE,DAVIANE M.C.; SANTANA,PATRÍCIA F.; ALMEIDA,ENRIK DE; COSTA,LUIZ P. DA; ALBUQUERQUE-JÚNIOR,RICARDO L.C. DE; LIMA-VERDE,ISABEL B.; PADILHA,FRANCINE F.; CARDOSO,JULIANA C..
ABSTRACT The objective of this work was to study the reduction in the capsaicin toxicity by encapsulation in liposomes. Capsaicin was extracted from peppers and characterized with high performance liquid chromatography (HPLC). We determined the zeta potential, polydispersivity index (PdI) and vesicle size of liposomes. Wistar rats were submitted to intravesical instillation of liposomes (LIP), capsaicin (CAP) or liposomes with capsaicin (CAPLIP). After 24 hours, bladders were removed for histological analysis. Vesicle size ranged from 68 to 105 nm with PdI smaller than 0.2 and zeta potential around -30 mV. The vesicles maintained stability over the 14-day study. The histological analysis of the CAP group showed intense inflammation in almost all bladder...

Diallel analyses and estimation of genetic parameters of hot pepper (Capsicum chinense Jacq.) Scientia Agricola
Sousa,João Alencar de; Maluf,Wilson Roberto.
The degree of heterosis in the genus Capsicum spp. is considered high; however, most of the studies refer to the species Capsicum annuum L. In spite of the potential use of F1 hybrids in pungent peppers of the species Capsicum chinense, few studies are available which assess the magnitude of heterosis in this species . This study was carried out to assess heterosis and its components in F1 hybrids from a diallel cross between hot pepper lines (Capsicum chinense) and to obtain data on the allelic interaction between the parents involved in the crosses. Trials were made in Rio Branco-Acre, Brazil, from March through October 1997. A randomized complete block design with fifteen treatments and three replications was used. The treatments were five C. chinense...

Expression and Distribution of Kinin B2 Receptor in Medulla and Spinal Cord Tissues of Rats Treated with Capsaicin International Journal of Morphology
de França,Camilla Estevão; Guardabassi,Carla; Smith,Ricardo Luiz.
Kinins are vasoactive peptides that promote pain and inflammation, yet centrally believed to participate to cardiovascular defensive reflexes produced by noxious stimuli. These peptides signal through the activation of two transmembrane G-protein-coupled receptors named B1 and B2 receptors (B1R and B2R). The B2R is constitutive in healthy tissues and animals. The aim of the study was to measure the gene and protein expression of B2R kinin receptors in central and peripheral tissues isolated from control rats and rats were pre-treated with capsaicin on the second day of life (50 mg/kg, s.c.) or two weeks prior to sacrifice (125 mg/kg over three days, s.c.). The same treatment with saline was made in control animals. Levels of mRNA for B2R were measured by...

Harvest seasons and pruning management in pepper: production and pungency of the fruits Horticultura Brasileira
Paulus,Dalva; Zorzzi,Ivan C; Rankrape,Fabiana; Paula,Fabiana M; Moura,Cláudia A.
ABSTRACT Changes in plant architecture, such as apical pruning and harvesting at the right time, are management practices to obtain not only greater fruit production, but mainly quality and pungent fruits, preferred by the consumer market. We evaluated the effect of management of pruning and harvest seasons on growth, production and pungency of fruits conducted in the field. Nine seasons of harvest were studied {90, 101, 116, 131, 146, 161, 176, 191, 206 and 221 days after transplanting (DAT)} and two management systems (pruning and no pruning). The treatments were arranged in a factorial design (9x2) with split plot in a randomized block design with three replications. The agronomic traits analyzed were: height, chlorophyll, leaf area, soluble solids,...

Inactivation of capsaicin-sensitive nerves reduces pulmonary remodeling in guinea pigs with chronic allergic pulmonary inflammation BJMBR
Prado,C.M.; da Rocha,G.Z.; Leick-Maldonado,E.A.; Starling,C.M.; Capelozzi,V.L.; Martins,M.A.; Tibério,I.F.L.C..
Pulmonary remodeling is an important feature of asthma physiopathology that can contribute to irreversible changes in lung function. Although neurokinins influence lung inflammation, their exact role in the extracellular matrix (ECM) remodeling remains to be determined. Our objective was to investigate whether inactivation of capsaicin-sensitive nerves modulates pulmonary ECM remodeling in animals with chronic lung inflammation. After 14 days of capsaicin (50 mg/kg, sc) or vehicle administration, male Hartley guinea pigs weighing 250-300 g were submitted to seven inhalations of increasing doses of ovalbumin (1, 2.5, and 5 mg/mL) or saline for 4 weeks. Seventy-two hours after the seventh inhalation, animals were anesthetized and mechanically ventilated and...

Involvement of the TRPV1 channel in the modulation of spontaneous locomotor activity, physical performance and physical exercise-induced physiological responses BJMBR
Hudson,A.S.R.; Kunstetter,A.C.; Damasceno,W.C.; Wanner,S.P..
Physical exercise triggers coordinated physiological responses to meet the augmented metabolic demand of contracting muscles. To provide adequate responses, the brain must receive sensory information about the physiological status of peripheral tissues and organs, such as changes in osmolality, temperature and pH. Most of the receptors involved in these afferent pathways express ion channels, including transient receptor potential (TRP) channels, which are usually activated by more than one type of stimulus and are therefore considered polymodal receptors. Among these TRP channels, the TRPV1 channel (transient receptor potential vanilloid type 1 or capsaicin receptor) has well-documented functions in the modulation of pain sensation and thermoregulatory...

Phenylpyrazolone derivatives inhibit gastric emptying in rats by a capsaicin-sensitive afferent pathway BJMBR
Vinagre,A.M.; Collares,E.F..
Dipyrone (Dp), 4-aminoantipyrine (AA) and antipyrine (At) administered iv and Dp administered icv delay gastric emptying (GE) in rats. The participation of capsaicin (Cps)-sensitive afferent fibers in this phenomenon was evaluated. Male Wistar rats were pretreated sc with Cps (50 mg/kg) or vehicle between the first and second day of life and both groups were submitted to the eye-wiping test. GE was determined in these animals at the age of 8/9 weeks (weight: 200-300 g). Ten minutes before the study, the animals of both groups were treated iv with Dp, AA or At (240 μmol/kg), or saline; or treated icv with Dp (4 μmol/animal) or saline. GE was determined 10 min after treatment by measuring % gastric retention (GR) of saline labeled with phenol red 10 min...
